Abstract:

A previously undiscovered transmembrane protein tyrosine phosphatase expressed only in bone and testis, osteotesticular protein tyrosine phosphatase (OST-PTP), has been identified, cloned, sequenced, expressed and subjected to enzymatic analysis. Also, a truncated osteoblast specific form (OST) containing the OST-PTP receptor but lacking the catalytic domain was identified and characterized. Based on the unique expression pattern of OST-PTP and OST during osteogenesis, assays can be used to screen for abnormal bone growth patterns and metabolic bone diseases. Moreover, this novel protein provides a specific target for use in conventional and gene therapies directed at treatment of osteoporosis, osteopetrosis and other bone metabolic disorders.

Live recombinant adenovirus containing a foreign gene coding for an antigen produced by another infectious organism is delivered to the intestine of a warm-blooded animal in an enteric-coated dosage form, whereupon the virus infects the gut wall and induces the production of antibodies or cell mediated immunity to both adenovirus and the other infectious organism.

Abstract:

There is disclosed a DNA transfer vector capable of replication in either a yeast cell or a bacterial cell and having inserted therein a DNA segment coding for human WA rotavirus surface antigen VP7. The transfer vector carrying the foreign DNA is used to transform yeast cells in order to produce the surface antigen VP7 which is capable of eliciting in vivo production of neutralizing antibodies with respect to human WA rotavirus. The surface antigen VP7 so elaborated and its use in a pharmaceutical vaccine composition are also disclosed.
