The present invention generally relates to a method for developing, generating and selecting tumor-free embryonic stem (ES)-like pluripotent cells using electroporation delivery of an inducible tumor suppressor mir-302 agent into mammalian cells. More particularly, the present invention relates to a method and composition for generating a Tet-On/Off recombinant transgene capable of expressing a manually re-designed mir-302 microRNA (miRNA)/shRNA agent under the control of doxycyclin (Dox) in human somatic/cancer cells and thus inducing certain specific gene silencing effects on the differentiation-associated genes and oncogenes of the cells, resulting in reprogramming the cells into an ES-like pluripotent state.

Abstract:
A system and a method of ensuring correct execution of a laboratory protocol or procedure are provided. An embodiment of the disclosure records videos of a bench area with a monitoring device, processes and stores the recorded video, and allows users to review the video. By reviewing the video of the lab procedure while conducting the laboratory protocol or procedure or shortly thereafter, users can resolve doubts and problems about particular steps of the lab procedure and conduct the procedure correctly.

Abstract:
A system for providing a form description language for a networked database program is disclosed. In one embodiment of the present invention, a database form is described using a plurality of components. Each component describes a particular item or entity within the database form. The size and placement of each of the various items in a database form is specified relative to the other items. The programming objects defining the structure of the database are created and stored separately from the data objects defining the data entered into the database by a user. A description language interpreter transmitted in conjunction with the database form allows display of the database form by executing the self-contained interpreter.
This invention is a deductive lockpicking puzzle, composed of a clamshell style frame holding a central cylinder that is representative in appearance to that of a normal lock, with a single rectangular keyway in the center. The four locking pins in this device each have two opposing magnetic poles, generated by attached permanent magnets, on either end. Each “lockpick” has a unique pattern of four magnets so that when inserted into the keyway, complementary magnetic poles will attract a pin to the lockpick, generating auditory and tactile feedback to the player upon impact. Once the right lockpick is inserted, all four locking pins are magnetically removed from their resting positions, allowing the central cylinder to rotate ninety degrees to a solved configuration.

Abstract:
This invention generally relates to a composition and method of using mam-made small RNAs, such as small interfering RNAs (siRNA), microRNAs (miRNA) and their hairpin-like precursors (pre-miRNA), as tumor suppressing anti-cancer drugs for treating human tumors and cancers, in particular, but not limited, for treating skin (melanoma), blood (leukemia), prostate, breast, liver and lung cancers as well as various neoplastic tumors, such as brain tumors and teratocarcinomas that contain a variety of tumorous and cancerous cells derived from all three germ layers of tissues, including ectoderm, mesoderm and endoderm. More specifically, the present invention relates to the use of miR-302-like siRNA (siR-302) and/or miR-302 precursors (pre-miR-302) for developing novel medicines and therapies against a variety of human cancers, in particular lung cancers.

Production And Utilization Of A Novel Anti-Cancer Drug In Therapy
This invention generally relates to a design and method for developing novel anti-tumor and/or anti-cancer drugs, vaccines and therapies, using microRNA and/or its shRNA homologues/mimics/derivatives. More specifically, the present invention relates to an use of a prokaryote-produced miRNA precursor (pro-miRNA) composition capable of being delivered into human cells and processed by the cells into mature miRNA effectors to elicit specific silencing effects on mir-302-targeted genes, subsequently leading to a beneficial result of tumor suppression and cancer therapy. The prokaryotic cells do not naturally express or process eukaryotic miRNA precursors (pre-miRNA); meanwhile, the present invention also teaches an inducible method for expressing pre-miRNAs, particularly mir-302 precursors by using the prokaryotic transcription system. Since mir-302 is a known tumor suppressor in human, this novel finding advances the design and method for developing new anti-cancer drugs, vaccines and/or therapies directed against multiple kinds of human tumors and cancers.
Novel Sugar Alcohol-Based Compositions For Delivering Nucleic Acid-Based Drugs In Vivo And In Vitro
This invention relates to a composition and its use for formulating nucleic acid-based drugs/vaccines with sugar alcohol compositions into complexes for both in-vitro and in-vivo delivery. Particularly, the present invention includes the ingredients and processes necessary for formulating therapeutic and pharmaceutical nucleic acid compositions, such as miRNA, microRNA precursors, shRNAs, siRNAs, ribozymes, antisense RNAs/DNAs, RNA-DNA hybrids and DNA vectors/vaccines, with glycylated sugar alcohols/sugars into delivery complexes, which can then be absorbed by cells in vivo and in vitro via active endocytosis. Also, the present invention discloses that chemical compounds containing sugar alcohol- and/or sugar-like structures can protect nucleic acids, in particular miRNAs, shRNAs, siRNAs and ribozymes, from degradation in vivo as well as in vitro. Therefore, the present invention is also a formula and method for preserving the structural integrity and functional efficacy of these nucleic acid-based drugs and/or vaccines in vivo and in vitro.
