Abstract:

The invention is directed to an isolated polypeptide which (a) is obtainable from a teleost; (b) has antimicrobial activity; (c) binds to oligoguanosine and/or CpG (SEQ ID NO:7); (d) comprises 58 strongly basic amino acids selected from the group consisting of K and R; (e) comprises 50 hydrophobic amino acids selected from the group consisting of A, I, L, F, W and V; (f) comprises 50 polar amino acids selected from the group consisting of N, C, Q, S, T and Y and (g) contains 11 lysine-rich motifs and antimicrobial fragments thereof as well as methods for preparing said polypeptides, compositions and libraries comprising said polypeptide(s) and uses of said polypeptide(s), particularly in treating microbial infections. The invention is further directed to a nucleic acid(s) encoding said polypeptide, microarrays comprising said nucleic acid(s) and uses for said nucleic acid(s). Furthermore, the invention is directed to an antibody which birds to the polypeptide of the present invention and uses for said antibodies.

Abstract:

The present invention encompasses a receptor molecule present on the surface of natural killer cells (NK cells) and non-specific cytotoxic cells (NCC) which is involved in non-specific lysis of target cells bearing an antigen recognized by the receptor molecule, an antigen common to the surface of cells recognized and lysed by natural killer cells, monoclonal and heterologous antibodies which bind to the receptor and target cell antigen(s) which are useful in their identification and purification, and methods for altering NK cell-mediated lysis of target cells. The monoclonal antibodies (mAbs) of the present invention were prepared by cell fusions between spleen cells from mice immunized with either non-specific cytotoxic cells (NCC) from catfish (anti-receptor antibodies) or NC-37 human lymphoblastoid B-cells, which are susceptible to lysis by NK cells, and myeloma cells. Eight distinct mAbs have been characterized, four directed against the NK/NCC target cell receptor and four against the NK/NC cell antigen. As used in the present invention "NK cells" will be used to include both natural killer cells of mammalian origin and non-specific cytotoxic cells of fish origin, unless specifically stated otherwise.

Abstract:

The present invention encompasses antigens recognized by the receptor protein, an antigen common to the surface of cells recognized and lysed by natural killer cells, monoclonal and heterologous antibodies which bind to the antigen proteins which are useful in their identification and purification, and methods for altering NK cell-mediated lysis of target cells. The monoclonal antibodies (mAbs) of the present invention were prepared by cell fusions between spleen cells from mice immunized with NC-37 human lymphoblastoid B-cells, which are susceptible to lysis by NK cells, and myeloma cells. The target cell antigen, a dimer of approximately 38,000 to 44,000 mw, seems to be evolutionarily conserved across species, from fish to mouse to man, an observation in agreement with the concept of NK cells being a primitive system, in terms of its appearance in evolution. The protein was purified from lysates of NC-37 cells by chromatography with mAbs bound to protein A beads followed by electrophoresis on 11% SDS-PAGE.

TM. with Con A Sepharose purified mAbs. Using mAbs which bind but do not inhibit lysis produces the same results as using mAbs which bind and inhibit lysis.

Abstract:

Utility of protein phosphatase inhibitors to protect teleost fish from microorganismic pathogens is disclosed. The present invention provides pharmaceutical compositions, kits, and methods of therapeutic and prophylactic treatments comprising sodium orthovanadate or vanadate-mimetic protein phosphatase inhibitors to protect fish, for example, catfish, from infection and disease caused by microorganismic pathogens, e. g. , Edwardsiella. Sodium orthovanadate, a protein phosphatase inhibitor, and vanadate-mimetic protein phosphatase inhibitors activate in vitro and in vivo the cytotoxicity of teleost nonspecific cytotoxic cells (NCC).
