The invention relates to the DNA and protein encoded by the GA4 locus. This protein is believed to be a member of the family of enzymes involved in the biosynthesis of the gibberellin family (GA) of plant growth hormones which promote various growth and developmental processes in higher plants, such as seed germination, stem elongation, flowering and fruiting. More specifically, the protein encoded by the GA4 locus is an hydroxylase. The invention also relates to vectors containing the DNA and the expression of the protein encoded by the DNA of the invention in a host cell. Additional aspects of the invention are drawn to host cells transformed with the DNA or antisense sequence of the invention, the use of such host cells for the maintenance, or expression or inhibition of expression of the DNA of the invention and to transgenic plants containing DNA of the invention. Finally, the invention also relates to the use of the protein encoded by the GA4 locus to alter aspects of plant growth.

Abstract:
The design of new ribozymes capable of self-catalyzed trans-splicing which are based upon the catalytic core of a Group I intron are described. Using this design, it is possible to construct ribozymes capable of efficiently splicing a new 3' exon sequence into any chosen target RNA sequence in a highly precise manner. A method of cell ablation is also described that provides a toxic product to a host cell in vivo in a targetted, regulated manner utilizing novel trans-splicing ribozymes of the invention. Inactive pro-ribozyme forms are also described.

Abstract:
A process for producing a transgenic mouse which contains and expresses the human insulin hormone gene. The insulin gene is expressed only in the pancreas of the transgenic mouse and is regulated by glucose, glucagon, or other insulin affectors in a manner which is indistinguishable from that of normal mice. Progeny of the trangenic mice inherit the human insulin gene in a Mendelian manner, with approximately 50% of the mice of each litter expressing the human insulin gene. The weights of the transgenic mice, growth rate, feeding behavior, reproductive capability, and longevity appear indistinguishable from normal mice. The mice are useful for studies of pharmacokinetics of insulin expression and for investigations of possible drug interactions with glucose homeostasis.
