Abstract:
The present invention relates to a method of detecting immune enhancement of virus infectivity, a method of determining neutralization and immune enhancement of virus infectivity, a method of identifying a virus epitope that displays immune enhancement, a method of identifying a compound that modulates activity of an Fc receptor, and a method of identifying a compound that modulates intracellular signaling of an Fc receptor. DNA constructs, cells, and kits relating to these assays are also disclosed.
Described herein are methods of improving the efficiency of gene transfer for a wide range of applications. Specifically provided are methods of increasing expression of an exogenous gene in a cell by contacting the cell with a vector comprising the exogenous gene and contacting the cell with a proteasome inhibitor, a lysosomal protease inhibitor and/or a microtubule inhibitor. Also provided are methods of delivering an antigen delivery vector to a cell or a subject. Provided are antigen delivery systems and kits comprising an antigen delivery vector and a proteasome inhibitor, a lysosomal protease and/or a microtuhulc inhibitor.

Abstract:
The disclosure provides a tetravalent Dengue virus vaccine, and methods of inducing a immune response against a Flavivirus such as Dengue virus 1-4 using the vaccine. The disclosure also provides methods of making a vaccine by introducing into a host cell a transgene that encodes a protein comprising Dengue domain III polypeptides, or, by purified dill polypeptides specific for Dengue serotypes 1-4, Yellow Fever Virus, West Nile Virus, Japanese Encephalitis Virus, and Flaviviruses.
