Proficient in using cell lines and primary cells, including tissue dissociation, ficoll/percoll cell separations, column and magnetic bead (MACS, Dynal) isolations. Immunologic assays: ELISpot, ELISA, antibody and neutralizing antibody titering, 51Cr release CTL, 3H incorporation (proliferation and suppression, MLRs, etc), dendritic cell, macrophage and long term lymphocyte culture and antigen-specific T cell line cloning, BLCL generation, in vitro immunization (IVI). Other in vitro assays/methods: cell based drug evaluations, histology, IHC, Multiplex (Luminex) assays, enzyme-substrate assays (SPM, -Gal,-Gal, SEAP, etc), XF bioenergenic assays, T-cell hybridoma and vaccinia vector generation, cloning, screening/purification, plaque assays, infections etc., Flow Cytometry (FACS): immunophenotyping (lymphocytes, Tregs, stem cells, alveolar macrophages, tumor and tumor stromal cells, etc.), MHC Class I tetramer staining, intracellular staining (transcription factors, cytokines, phospho-proteins), CFSE dilution, CFSE in vivo CTL assays, phagocytosis and ROS assays. Molecular Bio: immuno-blots, SDS-PAGE, gel electroporesis, plasmid preps, 32/33P in situ hybridization, transfections with electroporation, Fugene, calcium phosphate, etc., TREC RQ-PCR, organic extractions.
