Abstract:
The invention provides methods for predicting responsiveness of a subject to therapy with a CTLA-4 blocking agent. The methods involve assaying for at least one CTLA-4 polymorphism in the subject and predicting responsiveness of the subject to therapy with a CTLA-4 blocking agent based on presence or absence of a CTLA-4 polymorphic allele in the subject. The methods can further comprise selecting a treatment regimen with a CTLA-4 blocking agent in a subject based upon presence or absence of a CTLA-4 polymorphic allele in the subject. The methods can further comprise administering a CTLA-4 blocking agent to the subject according to the selected treatment regimen. Kits comprising a CTLA-4 blocking agent and means for assaying one or more CTLA-4 polymorphisms, optionally including a vaccine, are also provided.

Abstract:
A system and method for structure-based, small molecule activity prediction using binding mode prediction information. Binding scores between ligands and target molecules, (e.g. proteins, RNA, DNA, lipids, sugars) are first generated using molecular docking. A first machine learned deep neural network (DNN) model is developed using data representing the molecular ligand-target pair 3D structures and docking features to predict binding modes. Using transfer learning, weights of layers learned in the first machine learned model are used as weights in layers of a second machine learned DNN model used to more accurately improve the performance of activity prediction of the second machine learned model. For a target newly paired ligand-target complex, the method further implements a binding mode selector for selecting one or more particular binding poses for input to the activity prediction model for use in activity mode prediction of an activity of the target paired ligand-protein complex.
Target Molecule-Ligand Binding Mode Prediction Combining Deep Learning-Based Informatics With Molecular Docking
A computer-implemented method is described. The method includes generating, by a ligand bond graph generator, a first graph based on bond connectivity within a ligand molecule that is specified as input. The method further includes generating, by a ligand-protein graph generator, a second graph based on a contact map of the ligand molecule and a target molecule that is specified as another input. The method further includes receiving docking prediction metrics for the ligand molecule and the target molecule. The method further includes inputting, to a deep neural network, as input features, the first graph, the second graph, and the docking prediction metrics. The method further includes determining, using the deep neural network, a binding mode prediction that characterizes a set of potential interactions between the ligand molecule and the target molecule.

A multi-layer film and a film package made from the film are described herein that provide resealing capabilities by utilizing a tacky layer within the multi-layer film. The multi-layer film can include an outer film portion including the embedded tacky layer and an inner film portion. An opening feature formed in the multi-layer film includes a flap configured to be manipulated by a user to create an opening through the multi-layer film. The inner film portion can include a release layer configured specifically to interact with the tacky layer to provide a desired separation peel force and resealing functionalities. The outer film portion can include an outer film layer disposed on an opposite side of the tacky layer from the release layer that is configured to permanently adhere to the tacky layer.

Abstract:
A method is disclosed of predicting cancer patient response to immune checkpoint inhibitors, e.g., an antibody drug blocking ligand activation of programmed cell death 1 (PD-1) or CTLA4. The method includes obtaining mass spectrometry data from a blood-based sample of the patient, obtaining integrated intensity values in the mass spectrometry data of a multitude of pre-determined mass-spectral features; and operating on the mass spectral data with a programmed computer implementing a classifier. The classifier compares the integrated intensity values with feature values of a training set of class-labeled mass spectral data obtained from a multitude of melanoma patients with a classification algorithm and generates a class label for the sample. A class label “early” or the equivalent predicts the patient is likely to obtain relatively less benefit from the antibody drug and the class label “late” or the equivalent indicates the patient is likely to obtain relatively greater benefit from the antibody drug.

In one illustrative approach, the package opening feature is disposed adjacent or at a package seal, such as, for example, an end seal, a top seal, a bottom seal, a side seal, or a longitudinal seal. The flexible film laminate has at least first and second film layers, and at least one of the film layers defining an interior cavity configured to hold food products. In a number of configurations, the flexible film packages may be reclosable or resealable.
