Abstract:
The present invention provides a method of detecting a nuclease-mediated cleavage of a target nucleic acid through hybridizing a target nucleic acid to a fluorescently labeled oligonucleotide probe complementary to the target nucleic acid and containing a flourophor at one terminus and a quenching group at the other terminus. When the probe is unhybridized to the target nucleic acid, the probe adopts a conformation that places the flourophor and quencher in such proximity that the quencher quenches the flourescent signal of the flourophor and formation of the probe-target hybrid causes sufficient separation of the flourophor and quencher to reduce quenching of the flourescent signal of the flourophor. Once hybrized, the method contacts the probe-target hybrid with an agent having nuclease activity in an amount sufficient to selectively cleave the target nucleic acid and thereby release the intact probe. Detecting the release of the probe is then measured by following a decrease in the flourescent signal of the flourophor as compared to the signal of the probe-target hybrid.

Abstract:
A lace or draw string fastening system can include a clamping unit and a capture element. The clamping unit can clamp onto the laces, such as shoe laces, to tighten the shoes and the capture element can capture and contain the free ends of the laces that extend from the clamping unit to prevent tripping by securing them to other portions of the laces or the article that houses the laces. The fastening system can be used in a shoe lace fastening system without the need for knot tying. The clamping unit can clamp the laces together at the top of the shoe to hold the laces tight while the capture element can contain or secure the free ends of the shoe lace to the shoe or the laces adjacent the toe to prevent tripping.

Abstract:
An in vitro screening assay which identifies enzyme inhibitors and allows for the simultaneous assay of many enzymes. Enzyme, substrate, co-factor, etc. concentrations are optimized so that inhibitors of any one of the enzymes in the pathway are equally likely to be detected. Necessarily, the flux of substrate through each enzyme should be nearly the same during the assay, i. e. , each of the enzyme catalyzed steps must be equally rate-limiting. Preferably, optimal assay conditions are predicted by computer modeling. Further, the pathway conditions are optimized through variation of enzyme, starting substrate, co-substrate and co-factor concentrations. A positive response is initially detected as a change in the amount of the product generated at the end of the enzyme cascade as compared to a standard. A sample producing a positive result can be deconvoluted.
A method and system for providing access to aggregated data sets. The method includes organizing a plurality of data sets into an aggregated data set, providing access to at least a portion of the aggregated data set based upon a subscription level associated with a user, receiving a search query from the user to search the aggregated data, determining which portions of the aggregated data set the user can access based upon the subscription level of the user, filtering a set of search results based upon the determination of which portions of the aggregated data set the user can access, and returning the filtered search results to the user. Organizing the plurality of data sets into an aggregated data set includes receiving data from a plurality of sources, parsing the data, translating the parsed data into its native format and content, tagging the parsed data with attributes detailing how the data is entered, wherein the attributes comprise geographic location information, and mapping the translated data into a plurality of database tables within the aggregated data set. The system includes various components for performing the method.
