Abstract:
A novel collagen mimic comprising a tripeptide unit having the formula (flpYaaGly), where flp is 4(S)-fluoro-L-proline, is disclosed. The collagen mimic has increased stability relative to the collagen-related triple helices (ProYaaGly), (hypYaaGly), and (HypYaaGly).
Novel collagen mimics are disclosed with a tripeptide unit having the formula (Xaa-Yaa-Gly), where one of the positions Xaa or Yaa is a bulky, non-electron withdrawing proline derivative. By substituting a proline derivative at either the Xaa or Yaa position in the native collagen helix, the stability of the helix is increased due solely to steric effects relative to prior known collagen-related triple helices. Methods are also disclosed for making the novel collagen mimics.

Compositions and methods are provided for promoting bone growth. An implantable bone graft material is provided comprising a resorbable ceramic and a resorbable polymer, wherein the polymer comprises a covalently attached BMP binding peptide. In addition, an implantable bone graft material is provided consisting essentially of a resorbable β-TCP and a resorbable polymer, wherein the β-TCP has a total porosity of about 50% or greater and wherein the β-TCP has a particle size ranging from about 100 micron to about 300 micron. The implantable bone graft materials are useful for promoting bone growth in a subject.

The growth factor binding peptides include platelet derived growth factor (PDGF) binding peptides and growth differentiation factor (GDF) binding peptides. The tissue for repair includes tendon, muscle, connective tissue, ligament, cardiac tissue, vascular tissue, or dermis. Implantable devices for tissue repair are provided to which the cell and growth factor binding peptides are attached, such as acellular extracellular matrix having attached binding peptide.

In the methods, a sample containing cells is contacted with a cell binding peptide attached to a substrate, and the cells present in the sample are captured onto the substrate through binding to the cell binding peptide.
