Abstract:

A multiplex polymerase chain reaction assay that targets nine tetranucleotide short tandem repeat (STR) markers in the mouse genome. Unique profiles were obtained from seventy-two mouse samples that were used to determine the allele distribution for each STR marker. Correlations between allele fragment length and repeat number were determined with DNA Sanger sequencing. Genotypes for L929 and NIH3T3 cell lines were shown to be stable with increasing passage numbers as there were no significant differences in fragment length with samples of low passage when compared to high passage samples. In order to detect cell line contaminants, primers for two human STR markers were incorporated into the multiplex assay to facilitate detection of human and African green monkey DNA. This multiplex assay is the first of its kind to provide a unique STR profile for each individual mouse sample and can be used to authenticate mouse cell lines.

Abstract:

The present invention provides electrophoresis apparatus and electrophoresis methods employing gellan gum based gels employing divalent metal cation and diamine cross-linking agents. The gels are reversible under conditions that do not damage the biomolecules separated using the gels. The present invention also provides novel gellan gum-based gels which are cross-linked which employ a diamine cross-linking agent.

Abstract:

The invention provides fittings for the ends of electrochromatography cols. Each fitting includes an annular electrode surrounding one end of the passage electrical fields to be applied. The fitting also contains a passageway for eluant flowing into or out of the column, an annular chamber for the electrode and a membrane separating the passageway from the annular electrode chamber. The membrane prevents gases and electrolytic products from entering the column eluent going through the chromatographic column. The membrane also isolate the electrodes from the compounds such as proteins and macromolecules being separated. A buffer solution was pumped through the electrode chamber to remove the gases and electrode products. The buffer flow through the electrode chamber maintains a constant pH and conductivity so that the electrical field applied by each electrode is constant. The buffer flow also serves to remove the heat generated in the electrode chamber.

Abstract:

A method, kit and primers for determining alleles present in a DNA sample that is sourced from either a mouse or a cell line derived from a mouse. A set of STR marker loci are selected that can be amplified together in a multiplex amplification reaction. The set of STR marker loci are selected from the group consisting of: 18-3, 4-2, 6-7, 15-3, 6-4, 12-1, 5-5, X-1, Mo11-1, Mo19-2, Mo1-2, Mo7-1, Mo1-1, Mo3-2, Mo8-1, Mo2-1, Mo11-2, Mo17-2, Mo13-1, D8S1106 and D4S2408. A set of oligonucleotide primer pairs are provided. Each oligonucleotide primer pair flanks a single locus in the set of STR marker loci, which are co-amplified in a multiplex amplification reaction, with the product(s) comprising a mixture of amplified alleles from each of the co-amplified loci in the set of STR marker loci. The products are evaluated to determine the alleles present at each of the loci analyzed.
