Abstract:

A method for enhancing the conversion of a phenol substrate to a product by a peroxidase enzyme comprises the steps of reacting a conjugate comprising a detectably labeled phenol with a peroxidase enzyme in the presence of an enhancing reagent, the enhancing reagent conmprising an inorganic salt, an organic compound having the formula wherein when X is B(OH) , Y is I, or wherein when X is OH, Y is a halogen, or Q-R wherein Q is a linear or branched 1-12 heteroatom alkyl wherein the heteroatoms are selected from C, N, O, and S, wherein the bonds connecting the heteroatom alkyl chain are single or double, wherein any carbon atom in the heteroatom alkyl chain optionally includes a substituent selected from âOH, âCOOH, âNH , and âSH, and wherein R is selected from âOH, âCOOH, âNH , and âCH ; or mixtures of the inorganic salt and the organic compound. A compound having the formula: wherein R is âCH OH, âCOOH, and n is 1-8.

A method for performing a peroxidase-based assay by reacting a peroxidase enzyme with a soluble organic p-hydroxyphenyl-containing compound having the formula: wherein Q is a linear or branched 1-12 heteroatom alkyl wherein the heteroatoms are selected from C, N, O, and S, wherein the bonds connecting the heteroatom alkyl chain are single or double, wherein any carbon atom in the heteroatom alkyl chain optionally includes a substituent selected from âOH, âCOOH, âNH , and âSH, and wherein R is selected from âOH, âCOOH, âNH , and âCH ; and converting the soluble organic p-hydroxyphenyl-containing compound into a stable insoluble product which becomes highly fluorescent after illumination, upon excitation.

Abstract:

The present disclosure describes the use of macrolide antibiotics to prevent neurodegeneration and to treat or prevent disease and conditions which involve neurodegeneration. While not being limited to a specific mechanism of action, in one embodiment, the macrolide antibiotics inhibit neurodegeneration caused by lysosomal and/or mitochondrial dysfunction. The present disclosure contemplates the use of any macrolide antibiotic and pharmaceutically acceptable derivatives thereof. In a specific embodiment, the macrolide antibiotics include bafilomycin A1, bafilomycin B1 and concanamycin.

Abstract:

Novel peptides having gastrin releasing peptide-like activity having the structures Ala-Pro-Val-Ser-Val-Gly-Gly-Gly-Thr-Val-Leu-Ala-Lys-Met-Tyr-Pro-NH. sub. 2 and Gly-Asn-His-Trp-Ala-Val-Gly-His-Leu-Met-NH. sub. 2, have been isolated from rat and guinea pig brain extracts and synthesized by a solid phase peptide synthesis methods. The GRP-like peptides are useful as pharmaceuticals to lower body temperature, increase respiratory activity, reduce food intake, stimulate pancreatic hormone release, stimulate pituitary hormone release and suppress thyroid hormone release in mammals.
