The present invention provides methods and compositions for improving the efficiency of nucleic acid amplification reactions. The invention encompasses hybrid polymerases that show increased processivity over wild type polymerases as well as decreased exonuclease activity. The invention also encompasses methods, compositions and kits for conducting nucleic acid synthesis and amplification reactions in which non-specific amplification of primers is reduced.
Sso7-Polymerase Conjugates With Decreased Non-Specific Activity
The present invention provides methods for improving the specificity of nucleic acid amplification comprising incubating a nucleic acid molecule with a polymerase-Sso7 DNA binding domain conjugate and arginine, spermidine, or spermine. The present invention also provides reaction mixtures and kits for improving the specificity of nucleic acid amplification.
Nucleic Acid Detection By Bridge Amplification On Coded Particles
Particles that generate electromagnetic spectra upon appropriate illumination and in which the spectra can be converted to codes that reflect differences in the spectra are functionalized with a plurality of identical primer pairs on which bridge amplification can be performed. Using these particles, nucleic acids in a biological sample are amplified and exposed to a label that sends a signal only when bound to double-stranded DNA. The code for each particle is correlated with the primer pairs, and the captured nucleic acids are thus detected and identified. The procedure is useful for multiplex DNA or RNA detection as well as counting of target DNA and RNA molecules in the sample.

Abstract:
Provided are methods for detecting the presence of multiple barcoded solid supports in partitions, the method comprising providing a plurality of partitions wherein at least some partitions comprises multiple solid supports, where each solid support is linked to different solid support oligonucleotides, the solid support oligonucleotides comprising a barcode unique for the solid support; synthesizing a complementary sequence of the oligonucleotides to produce a double-stranded polynucleotide having barcodes from two of the solid support oligonucleotides; and sequencing one or both strands of the double-stranded polynucleotides, wherein sequences comprising two different barcodes indicates the presence of two solid supports in the same partition. Also provided are compositions and methods for producing the oligonucleotides.
