Abstract:

The overexpression of certain marker genes including Wnt5a has been found useful in the identification of more aggressive forms of malignant melanoma. Therefore, the overexpression of these genes in tumor samples of malignant melanoma may be useful in the diagnosis, profiling, and treatment of patients suffering from this disease. Inhibitors of Wnt5a activity may be useful in the treatment of aggressive forms of malignant melanoma. Inhibition of Wnt5a activity may be effected by any method including anti-sense therapy, gene therapy, and pharmaceutical intervention.

Abstract:

Methods for assaying a sample for a probe polynucleotide are provided. The methods comprise forming a complex between a target on a substrate, the probe polynucleotide that binds to the target, and a conjugate comprising a semiconductor nanocrystal that binds to the probe polynucleotide by way of a tag sequence on the probe polynucleotide. The complex is formed when the probe polynucleotide is present in the sample. The methods are useful in any technique in which the detection of a target that can bind to a probe polynucleotide is desired, for example in fluorescence in situ hybridization. The methods are particularly useful in multiplex settings such as hybridization to microarrays where a plurality of targets are present. Assay complexes produced by such methods and kits useful for performing such methods are also provided.

Abstract:

Gene expression can be quantitatively analyzed by hybridizing fluor-tagged mRNA to targets on a cDNA micro-array. Comparison of gene expression levels arising from co-hybridized samples is achieved by taking ratios of average expression levels for individual genes. In an image-processing phase, a method of image segmentation identifies cDNA target sites in a cDNA micro-array image. The resulting cDNA target sites are analyzed based on a hypothesis test and confidence interval to quantify the significance of observed differences in expression ratios. In particular, the probability density of the ratio and the maximum-likelihood estimator for the distribution are derived, and an iterative procedure for signal calibration is developed.
