Abstract:

The invention relates to a method of stimulating antibody-dependent cellular cytotoxicity to enhance the elimination of IgE-bearing B-cells comprising administering to a mammal an anti-IgE antibody, which binds to membrane bound IgE, but does not induce histamine release and administering an ISO to the mammal. The ISO may be a CpG containing oligonucleotide, or a modified CpG-containing oligonucleotide with an electron-withdrawing group at least at position C-5 of the cytosine in the CpG sequence. In addition, the method may include the administration of an allergen to improve desensitization therapy.

Abstract:

The invention is related to conjugates comprised of a pro-apoptotic protein encoded by Bak, Bak-2 or Bax genes and a ligand specifically targeting a cell surface antigen. The cell surface antigen is expressed on tumor cells, cells infected with bacteria or virus, or cells of specific functions, but it is not expressed or expressed only to a limited extent on other cell types. Abstract:

A method of determining the antigens encoded by a genomic or cDNA library is disclosed. Dendritic or other antigen presenting cells are transfected with DNA fragments in a vector which includes a signal peptide coding sequence and an sequence which encodes a peptide binding to a receptor on the antigen presenting cell. The expressed DNA fragments are secreted under control of the signal peptide, and bind to a cell surface receptor. The antigen presenting cells are used to generate monoclonal antibodies. The monoclonal antibodies may be screened by cloning the same fragments into a display vector containing a transmembrane domain thereby displaying the expressed proteins on the surface of a host cell. The monoclonal antibodies are screened against these displayed proteins for a positive match.
