Abstract:
Described are microfluidic devices for, preferably, high-throughput, multi-analyte, affinity capture and detection of affinity-bindable analytes in biological fluids. Particularly, the devices can be used for immunoassays of biological fluids using multiple antibodies for capture and detection of multiple analytes, including proteins. The devices can be used for the simultaneous isolation and quantization of multiple proteins from microliter samples of biological fluids. Also described are methods for detecting and, optionally, quantifying, affinity-bindable analytes in biological fluids using these devices.

A fixed-optics system for detecting fluorescence light emitted from one or more spatially selected volumes (S), e.g., in one or more microfluidic channels, comprises a detection optical fiber () corresponding to each selected volume and a ball lens () adjacent to one end of each optical fiber, wherein the ball lens has a diameter (D) larger than the diameter of the optical fiber and a distance (d) between the ball lens and the optical fiber is in a range of 3 to 40 times the diameter of the optical fiber, so that due to a quasi-focusing configuration light emitted from the spatially selected volume and received by the ball lens converges onto the end () of the optical fiber.

Abstract:
Embodiments of a microfluidic system and method for stimulating a blast shock wave that supports a short and defined laminar flow of a liquid media solution through a microfluidic device such that sheer stress on neural tissue disposed within the microfluidic device is precisely controlled are disclosed. The microfluidic system includes a pneumatic device applies a blast shock wave having a quick rise time across a microfluidic channel of the microfluidic device. The microfluidic device includes an inlet reservoir in fluid flow communication with an outlet reservoir through the microfluidic channel. The inlet and outlet reservoirs are secured to a top structure which is attached to a bottom structure that collectively defines the microfluidic channel with a cover slip that is attached to the underside of the bottom structure.

The present disclosure is directed to embodiments of microstructured membranes, methods of fabricating microstructured membranes, bioreactors housing microstructured membranes, and methods of using bioreactors and microstructured membranes. In some embodiments, the present disclosure allows culturing of cellular tissues in an environment which more accurately resembles a native environment. In some more specific embodiments, the present disclosure allows culturing of tumor cells on a membrane having a microfabricated pattern which mimics a native vasculature system.
