Weight Loss Induced By Reduction In Neuropeptide Y Level
Thomas Loftus - Great Falls VA, US Craig Townsend - Baltimore MD, US Gabriele Ronnett - Lutherville MD, US M. Lane - Baltimore MD, US Francis Kuhajda - Lutherville MD, US
International Classification:
A61K031/365
US Classification:
514/473000
Abstract:
This invention provides a method for inducing weight loss in an animal by administering to the animal a compound which reduces the expression and/or secretion of neuropeptide Y (NPY). The effect may be accomplished directly, indirectly or humorally. Preferably, administration of this compound has the effect of increasing malonyl CoA levels in the animal. Compounds administered according to this invention may be inhibitors of fatty acid synthase (FAS), including substituted -methylene--carboxyl--butyrolactones, or inhibitors of malonyl Coenzyme A decarboxylase (MCD). Preferably, the compound is administered in an amount sufficient to reduce the amount and/or duration of expression and/or secretion of NPY to levels at or below those observed for lean animals. In another preferred embodiment, the administration will reduce expression and/or secretion to levels observed for fed or satiated animals; more preferably, administration will reduce the level of NPY below that of fed animals. In a particular embodiment, this invention provides a method for inducing weight loss in an animal by administering a compound which inhibits feeding behavior in the animal. The method is particularly useful for inducing weight loss in animals deficient in expression of the hormone leptin or animals resistant to the action of leptin.

The effect may be accomplished directly, indirectly, or humorally. Preferably, administration of this compound has the effect of increasing malonyl CoA levels in the animal. Compounds administered according to this invention may be inhibitors of fatty acid synthase (FAS), including substituted α-methylene-β-carboxyl-γ-butyrolactones, or inhibitors of malonyl Coenzyme A decarboxylase (MCD). Preferably, the compound is administered in an amount sufficient to reduce the amount and/or duration of expression and/or secretion of NPY to levels at or below those observed for lean animals. Abstract:
An atomic oscillator device includes an atomic oscillator, a controlled oscillator, a resonance controller, and a cold-atom clock output. The atomic oscillator comprises a two-dimensional optical cooling region (2D OCR) for providing a source of atoms and a three-dimensional optical cooling region (3D OCR) for cooling and/or trapping the atoms emitted by the 2D OCR. The atomic oscillator comprises a microwave cavity surrounding the 3D OCR for exciting an atomic resonance. The controlled oscillator produces an output frequency. The resonance controller is for steering the output frequency of the controlled oscillator based on the output frequency and the atomic resonance as measured using an atomic resonance measurement. The cold-atom clock output is configured as being the output frequency of the controlled oscillator.
