Abstract:

The present invention is a method for ensuring client access to paired and unpaired response messages. In the method, the server detects one or more unpaired message which are stored in a data structure on the server. The data structure may be created when the server is started or automatically when the first unpaired message is identified. The method then uses a communications protocol between the client and server which allows the client to request one or more of the stored unpaired messages. The present invention uses a request module configured to receive a client request and prepare the client request for a response generator. The response generator receives the client request from the request module and generates an appropriate response. Next, an unpaired message module analyzes the response message generated by the response generator and distinguishes a paired message from an unpaired message. Then, the unpaired message module stores paired messages in an unpaired response data structure.

Abstract:

Abstract:

Methods of creating libraries of biological polymers are provided. The construction of a library employs a probability matrix for a reference sequence, and a constraint vector for which is applied to the probability matrix to produce a substitution scheme. The substitution scheme is then used to generate a library comprising substitutions recommended by the substitution scheme. The library members, or host cells comprising and/or expressing them, can be screened for desired changes in a property of interest in the biological polymers in the library.

Abstract:

A high throughput mutagenesis screening method selects sites for saturation scanning using protein structural considerations. Site-saturation libraries are created and screened using assays for sites having protein properties of interest. The sites are categorized for the properties of interest, trends, if any, are identified; and, a variant having a desired property is selected for additional library creation procedures. The additional libraries are created using feedback from the determined categories. One set of additional libraries repeats construction of the previous libraries using the feedback. A second set of additional libraries are new libraries created at sites that are expected to be beneficial based on the feedback. The categories may be coded, using color or other indicia, to allow easy identification of trends.
