Abstract:
Modified thermostable DNA polymerases having enhanced efficiency for incorporating unconventional nucleotides such as those labeled with fluorescein family dyes are advantageous in many in vitro DNA synthesis applications. Such enzymes are particularly useful for use in chain termination nucleic acid sequencing protocols, as are native forms of such enzymes. Genes encoding the modified enzymes and methods for their production and use offer cost and efficiency advantages for DNA sequencing.
Crosspoint Microwave Switching Architecture For Optical Telecommunications
The present invention combines several features to create a novel type of optical telecommunications switching component using microwave waveguides as opposed to the usual methods employing light deflection or digital signal processing. Some of the components of the device include: (1) an optical crossconnect (OXC) which allows any combination of wavelengths to be added or dropped from an element to an optical network, (2) an optical add/drop multiplexer (OADM) which allows any wavelength to be converted to any other, (3) a microwave crossbar switch array optimized for this purpose, and (4) a means for reshaping the exiting signal from the microwave crosspoint switch array without regard to signal format and without need for complex and expensive digital signal processing. The minimal architecture for this functionality is shown to be three NÃN microwave crossbar switch arrays.
Integrated Fifo Memory Management Control System Using A Credit Value
An integrated data controller that utilizes a first-in first-out (FIFO) management system that compensates for the unpredictable nature of latency associated with requesting data from memory and enables the timing of data requests to be determined based on the number of pending requests and the amount of data currently residing in the buffer. The FIFO management system includes a FIFO controller and a FIFO buffer that monitor a credit value and a trigger value to determine when to make data request bursts upon a memory unit. The trigger value is an indication of whether there is a sufficient amount of free space for it to be beneficial to make a data request burst and the credit value is a number that indicates the number of a data blocks that should be requested in the data request burst.

The present invention provides thermostable or thermoactive DNA polymerases with attenuated 3′-5′ exonuclease activity, methods for their synthesis, methods for their use, kits comprising the polymerases, nucleic acids encoding the polymerases and cells comprising such a nucleic acid. A programmable data filtering/searching system for use with a data network. The primary programmable hardware-based filtering and searching portions of the system are integrated onto the same printed circuit board as the data storage device to facilitate efficient filtering and searching of data from a data stream coming from a memory storage device after being captured. In addition, the actual hardware and software used to perform the filtering and searching routines is programmable such that various filter and search criteria can be entered and utilized. Continuously Providing Instructions To A Programmable Device
A system and method for enabling a programmable device to execute instructions without interruption. An instruction space for storing instructions from a host application is bifurcated to define a program segment and a hold segment. At startup, instructions are loaded into the hold segment, and the programmable device begins executing those instructions. While the hold segment instructions are executed, the program segment is loaded with instructions. Once the program segment is filled, control is shifted to it and instructions from this segment are executed by the programmable device. When the program segment has been executed, control is shifted back to the hold segment, and instructions are taken from it while the program segment is reloaded with a fresh set of instructions from the host application. Once the program segment is reloaded, control is redirected and execution of instructions from the program segment is continued.
