Abstract:
The present invention is generally related to compositions, kits and methods for labeling nucleic acid molecules using reverse transcriptases, preferably multi-subunit reverse transcriptases such as ASLV reverse transcriptases. Specifically, the invention relates to methods, kits and compositions for fluorescently labeling nucleic acid molecules during nucleic acid synthesis. The labeled nucleic acid molecules produced in accordance with the invention are particularly suited as labeled probes for nucleic acid detection and diagnostics.

The present invention generally relates to methods of making cDNA molecules and cDNA libraries. The invention also relates to cDNA molecules and cDNA libraries produced according to these methods, as well as to vectors and host cells containing such cDNA molecules and libraries. The invention also relates to kits for making the cDNA molecules and libraries of the invention.
The present invention provides a method for the rapid isolation and recovery of a desired target DNA or RNA molecules from a mixture or library containing such molecules. The method involves the use of biotinylated probes and enzymatic repair-cleavage to eliminate undesired library members from a sample.

International Classification:
C12Q001/68
US Classification:
435/091100, 435/006000, 435/091200
Abstract:
The subject invention provides novel compositions containing one or more peptides or polypeptides which possess ribonuclease activity. Preferably the ribonucleases for inclusion in the compositions are ribonucleases that substantially lack deoxyribonuclease activity. A preferred embodiment is a composition comprising RNase A or RNase T1. Another aspect of the invention is to provide methods for synthesizing nucleic acids, typically DNA, using the compositions of the invention or one or more ribonucleases. Another aspect of the invention involves the use of the subject method of nucleic acid synthesis to carry out the synthesis step in a polymerase chain reaction experiment. Yet another aspect of the invention is to provide kits for the synthesis of nucleic acids, wherein the kits comprise one or more peptides or polypeptides that possess ribonuclease activity.

Abstract:
The present invention is directed to methods for the production and isolation of nucleic acid molecules. In particular, the invention concerns isolation of mRNA molecules and the production and isolation of nucleic acid molecules (e.g., cDNA molecules or libraries), which may be single- or double-stranded. Additionally, the invention concerns selection and isolation of particular nucleic acid molecules of interest from a sample which may contain a population of molecules. Specifically the invention concerns affinity-labeled primer-adapter molecules which allow improved isolation and production of such nucleic acid molecules, increasing both product recovery and speed of isolation.
