Molecular Biology: primer design, PCR techniques (RT-PCR, overlap-extension PCR, site-directed mutagenesis); RNA extraction; generation of cDNA constructs encoding fused recombinant proteins and secreted recombinant proteins for expression in Prokaryotic, Yeast or Mammalian expression systems; preparation of chemically competent Escherichia coli and Pichia pastoris cells; E. coli and Pichia pastoris transformation; generation of bicistronic co-expression vectors Protein/enzyme purification, identification and assessment of physical properties, functional conformation and activity: Protein expression (Escherichia coli, Pichia pastoris) and purification applying a variety of chromatographic methods (FPLC-Akta system; HPLC); protease assays using natural substrates or synthetic substrates; protease inhibition assays; active-site titration assays; Fluorescence Spectroscopy (fluorescent probes; anisotropy) and Mass Spectrometry (LC-MS, MALDI-TOF); SDS-PAGE, Western-blot, immunoprecipitation, Co-IP, immunoblotting, biotinylation; Interferometry (BLI-Octet) Experience in enzyme kinetics in order to understand and apply the available technologies Experience with assay development using ELISA and Multiplex Technology, among others Demonstrated success with the development of High Throughput screening assays Good working knowledge of Microsoft suite of Software (Word, Excel), Python, OriginLab, Vector NTI and protein structure analysis programs.
