Abstract:

A polymeric matrix for delivery of an HMG CoA reductase inhibitor such as a statin to tissue such as cardiac tissue in need thereof for the treatment or prevention of a disease or defect such as atrial fibrillation has been developed. In the preferred embodiment, a statin is delivered by means of a patch sutured to cardiac tissue at the time of cardiothoracic surgery. In the most preferred embodiment, the patch is a biodegradable material providing controlled or sustained release over a prolonged period of time, such as a week. Suitable materials include extracellular matrix, or other biodegradable hydrogels or polymeric materials providing sustained or controlled release of statin at the site of application.

Abstract:

Pharmaceutical formulations and methods of using thereof for the treatment and prevention of pulmonary arterial hypertension are provided. The formulations contain one or more agents to simultaneously reduce ADMA levels in a patient and reduce inflammatory processes in the pulmonary vasculature of a patient. The formulations contain a therapeutically effective amount of cerivastatin, a cerivastatin analog, or a pharmaceutically acceptable salt, prodrug, clathrate, or solvate thereof in a carrier suitable for pulmonary administration.

Abstract:

Compositions and methods are provided for metabolically degrading ADMA. In one embodiment a device is provided for reducing a patients ADMA levels in their blood wherein the device comprises a biologically active dimethylarginine dimethylaminohydrolase (DDAH) polypeptide covalently linked to a solid support. In one embodiment a method for reducing ADMA levels in a patients blood comprises the step of contacting the patients blood or a blood fraction with an immobilized biologically active DDAH polypeptide, wherein contact of the patients blood with said DDAH polypeptide results in degradation of ADMA present in the patients blood.
