Abstract:
A traffic management system for an automatic material handling system (AMHS) divides time into a series of discrete time periods and analyzes a plurality of move requests that are to be executed within a particular time period is disclosed. The traffic management system receives a plurality of move requests from the AMHS and determines how many move requests are to be executed and selects which move requests in particular will be executed in the next time period. The traffic management system prioritizes the selected move requests by analyzing each source node and destination node contained in a move request in conjunction with other node traffic data. The traffic management system analyzes this data to ensure that a move request is not executed if a material transport vehicle would be dispatched to a node having a node traffic balance value that exceeds a predetermined value and a node traffic density value that exceeds a predetermined value.

Abstract:
Synthesis of many proteins is tightly controlled at the level of translation and plays an essential role in fundamental processes such as cell growth and proliferation, signaling, differentiation or death. Methods that allow imaging and identification of nascent proteins allow for dissecting regulation of translation, both spatially and temporally, including in whole organisms. Described herein are robust chemical methods for imaging and affinity-purifying nascent polypeptides in cells and in animals, based on puromycin analogs. Puromycin analogs of the present invention form covalent conjugates with nascent polypeptide chains, which are rapidly turned over by the proteasome and can be visualized and specifically captured by a bioorthogonal reaction (e.g., [3+2]cycloaddition). The methods of the present invention have broad applicability for imaging protein synthesis and for identifying proteins synthesized under various physiological and pathological conditions in vivo.

Abstract:
By hijacking endogenous E3 ligase to degrade protein targets via the ubiquitin-proteasome system, PROTACs (PRoteolysis TArgeting Chimeras) provide a new strategy to inhibit protein targets that were previously regarded as undruggable. The compounds described herein comprise a photolabile group on PROTACs, enabling the degradation of protein targets in a spatiotemporally controlled manner By adding a photolabile caging group on ubiquitin recruiting moieties, light-inducible protein degradation was acheived. These opto-PROTACs display no activity in the dark, while restricted degradation can be induced at a specific time and rate by UVA-irradiation. Accordingly, these compounds provide light-controlled PROTACs and methods of using such compounds.
