Provided herein are amyloid-binding assays that simulate in situ brain conditions by combining binders with various A-beta species on mammalian brain tissue. In general the amyloid-binding assays comprise the steps of (a) contacting a mammalian brain tissue sample with at least one A-beta species; (b) applying a putative binder to the brain tissue sample; and (c) determining whether the putative binder binds to the A-beta species.

The present invention relates to methods for the detection of myelin and a quantitative measurement of its local concentration in a sample using a benzofuran compound or its radioisotope derivatives. In one embodiment a method of myelin detection comprises identifying a subject at risk of or diagnosed with a myelin-associated neuropathy, parenterally administering to the subject the benzofuran compound, or a derivative of the compound, and determining myelination in the subject by detecting binding in the sample. In one embodiment, the invention provides a method of imaging myelinated tissue in a surgical field of mammalian tissue comprising the steps of contacting the surgical site with the benzofuran compound or a radioisotope derivative of the compound, and detecting binding in the surgical site. In yet another embodiment, a method of imaging spinal cord and spinal nerve root tissue is provided comprising the steps of parenteral administration of a radioisotope derivative of the benzofuran compound and detecting the radioisotope derivative within the spinal canal and intervertebral foramen. Methods of quantification of binding of the benzofuran compound or its derivatives are also provided.

Improved methods for forming substituted polymers having a high degree of conjugation include the steps of activating a steric hindrance molecule under conditions that ensure substantially all the steric hindrance molecules are mono-activated and then reacting with a polymer. An improved method for recovering polypeptides substituted with steric hindrance molecules and having an elongate configuration removes impurities and volatiles without achieving complete dryness of the copolymer.

Abstract:
The present disclosure generally relates to systems and methods for identifying the boundaries of tumors and assessing quantitatively the ability of dyes to highlight a tumor's boundary. In accordance with these methods and systems, images are taken of subjects administered agents labeled with dyes. After accessing the images, tumors are selected and routines employed to both identify the boundaries of the tumors, as well as, to quantify various aspects of the tumor boundaries. From these quantifiable descriptors the performances of the various dyes to highlight the boundaries of tumors are evaluated.
