Abstract:
The present invention provides compositions and methods for research, diagnostic, drug screening, and therapeutic applications related to colorectal cancer and related conditions. In particular, the present invention provides genetic variations in or associated with one or more of the STK38L, GPR45, TGFBRAP1, PADI3, IBRDC1, and/or GRK5 genes as being associated with such conditions.

Methods are provided herein to determine if a subject is a candidate for treatment with an inhibitor of 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGCR). The method includes determining the presence of at least one polymorphism in the HMGCR gene in a sample from a subject. The presence of at least one polymorphism indicates that the subject is a candidate for treatment with a statin, for example to decrease risk of or treat cancer, cardiovascular disease, diabetes, obesity, inflammatory disease, or auto-immune disease.

During routine screening of a patient with a family history of colorectal cancer for truncating APC mutations, a novel missense mutation was identified. Upon further evaluation, it was found that 6% of Ashkenazi Jews carry this mutation, and that it was present in. sup. about. 20% of Ashkenazis with a family history of CRC. Probes, methods, and kits for identifying individuals affected with this missense mutation are provided.
